Genetic Mutation Signature for Relapse Prediction in Normal Karyotype Acute Myeloid Leukemia

چکیده
Background: Risk stratification for normal karyotype acute myeloid leukemia (NK-AML) remains unsatisfactory owing to the high incidence of relapse. This study aimed evaluate role gene mutations and clinical characterization in predicting relapse patients with NK-AML. Methods: A prognostic system NK-AML was constructed based on mutations, measurable residual disease (MRD), characteristics. panel explored using next-generation sequencing. The least absolute shrinkage selection operator, nomogram algorithm were used build a genomic mutation signature (GMS) (GMSN) model that combines GMS, MRD, factors predict 347 from four centers. Results: Patients GMS-high had higher 5-year than those GMS-low (P < 0.001). also GMSN-high GMSN-intermediate -low disease-free survival overall rates lower 0.001) as confirmed by training validation cohorts. Conclusions: illustrated potential GMSN predictor

Genetic Risk Prediction for Normal-Karyotype Acute Myeloid Leukemia Using Whole-Exome Sequencing
Normal-karyotype acute myeloid leukemia (NK-AML) is a highly malignant and cytogenetically heterogeneous hematologic cancer. We searched for somatic mutations from 10 pairs of tumor and normal cells by using a highly efficient and reliable analysis workflow for whole-exome sequencing data and performed association tests between the NK-AML and somatic mutations. We identified 21 nonsynonymous si...
متن کاملAn mRNA expression signature for prognostication in de novo acute myeloid leukemia patients with normal karyotype.
Although clinical features, cytogenetics, and mutations are widely used to predict prognosis in patients with acute myeloid leukemia (AML), further refinement of risk stratification is necessary for optimal treatment, especially in cytogenetically normal (CN) patients. We sought to generate a simple gene expression signature as a predictor of clinical outcome through analyzing the mRNA arrays o...
متن کاملPrognostic significance of the FLT3 ITD mutation in patients with normal-karyotype acute myeloid leukemia in relapse
BACKGROUND Fms-like tyrosine kinase 3 internal tandem duplication (FLT3 ITD) mutation is related to poor prognosis in normal-karyotype acute myeloid leukemia (AML). However, the prognostic significance of the mutation at relapse has not been adequately investigated. We investigated the prognostic significance of the FLT3 ITD mutation at relapse in normal-karyotype AML patients. METHODS We ana...

New Fusion Transcripts Identified in Normal Karyotype Acute Myeloid Leukemia
Genetic aberrations contribute to acute myeloid leukemia (AML). However, half of AML cases do not contain the well-known aberrations detectable mostly by cytogenetic analysis, and these cases are classified as normal karyotype AML. Different outcomes of normal karyotype AML suggest that this subgroup of AML could be genetically heterogeneous. But lack of genetic markers makes it difficult to fu...
